Title: Engineered probiotics for detecting C. difficile infection and delivering nanobody-based therapeutics. Description: Antibiotics disrupt gut microbiota, allowing C. difficile to bloom, produce toxins, and cause recurrent diarrhea. Current treatments risk further dysbiosis, demanding smarter solutions. C. difficile releases p‑cresol, a unique metabolite that can serve as a detection signal. We engineered probiotic E. coli Nissle 1917 with a p‑cresol sensor (from Aromatoleum aromaticum) that activates GFP fluorescence, converting chemical cues into real‑time visible signals. Beyond detection, our engineered strain is designed to release therapeutic nanobodies on demand to neutralize C. difficile toxins, helping restore microbiota balance. By enabling probiotics to sense and respond, we aim to build safer, smarter tools against recurrent CDI. We are the iGEM team from Shimen High School, China. Official Page
