<description>Title: FusionDrive: Engineering and Functional Evaluation of an Anti-TIGIT–IL-2v Fusion Protein for Gastric Cancer Immunotherap Description: Dietary habits, Helicobacter pylori infection, smoking, and alcohol exposure contribute to Shandong’s high gastric cancer burden. Treatments remain limited by chemoradiotherapy toxicity, low anti-PD-1 response rates, and IL-2-associated systemic toxicity and off-target immunosuppression. We designed FusionDrive, fusing an anti-TIGIT antibody with an engineered IL-2 variant. Across five engineering cycles, we expressed and purified proteins in CHO cells and evaluated target binding and signaling activity. The variant retained TIGIT binding (EC50: 1.155 nM). In a PHA-induced Jurkat model, it reduced pSTAT5 signaling more strongly in CD25⁺ cells than in CD25⁻ cells relative to the wild-type fusion, while retaining CD25⁻ signaling activity, supporting reduced CD25-associated response bias. Human Practices integrated stakeholder feedback into project iteration, while our business plan outlined a translation pathway guided by research milestones and industry collaboration.
