Title: PARCEL: Pumilio-Assisted RNA Carrier via ELP Transition for Asthma Treatment Description: The prevalence of asthma is increasing worldwide. Caused by genetic and environmental factors, asthma can severely impair respiratory function and patients' quality of life, yet existing treatments have limitations. We developed PARCEL: Pumilio-Assisted RNA Carrier via ELP Transition for Asthma Treatment, utilizing phase separation of elastin-like polypeptide (ELP) to build protein-mRNA supermolecules for inhaled asthma medication. Integrated ELP-Pum2-EEP13 fusion proteins assemble into micellar nanoparticles, with therapeutic mRNAs captured inside by the Pumilio 2 (Pum2) domain. Via Sortase A-induced antibody conjugation, the nanoparticles can specifically target lung epithelial cells and disassemble after internalization. Triggered by endosomal escape peptide 13 (EEP13), RNAs are released to be translated into asthma-related proteins. Our design establishes a novel RNA delivery platform, throwing new light on safe and specific therapies for asthma and beyond. Wiki | Official Page
