Title: ProLac: Characterization of a Probiotic and In Silico Optimize Lactose Consummption for Lactose Intolerance Relief Description: Lactose intolerance is a highly prevalent digestive disorder. Over 90% of adults in Asia lose lactase persistence, and many of these people suffer from symptomatic lactose intolerance. Distinct from previous iGEM teams that aimed to construct new engineered strains, we launched ProLac, an observation-first synthetic biology project. We characterized an acid- and oxygen-tolerant probiotic Lacticaseibacillus paracasei subsp. paracasei CICC 6109 with growth curve assays and plasmids sequence analysis. With COBRApy, we performed in silico manM knockout simulation on Lacticaseibacillus paracasei ATCC 334 based on a published genome-scale metabolic model to redirect carbon metabolism for preferential lactose utilization. To address the wet-lab experimental challenges, we managed to develop a multifunctional fluorometer. We also built an open-source tool for campus biology map for science outreach and released our campus map as a demo. Wiki | Official Page
