Title: BLISS: Bioengineered Low-calorie Innovative Sweetener Synthesis Description: Current sugar substitutes often compromise taste and deprive people joy of eating. The sweet protein Brazzein, from Pentadiplandra brazzeana, combines a clean, sugar-like taste with promising stability during processing. However, difficulties with protein folding and purification in recombinant microbial expressions limit its production for broader commercial use. BLISS addresses these challenges through a novel active inclusion body strategy in Escherichia coli. Using the SAP peptide (L6KD) and partner adapter tags (Sumo and MtuI), we demonstrated improved Brazzein production and simplified its recovery. With AI-assisted protein engineering, we identified variants with potentially better stability and sweetness. We also evaluated expression in yeast for scalable, food-grade production. Through iterative engineering, we develop PrintScope, a figure-sized miniature microscope, to promote accessible biology education. Ultimately, BLISS seeks to create a sweet future without the associated health burden. Wiki | Official Page
