Title: OrbitColi Description: During long-term space missions like Artemis, astronauts are exposed to cosmic radiation and microgravity, leading to oxidative stress that conventional antioxidant supplements have limited efficacy due to instability and fixed dosing. We designed OrbitColi, an engineered Escherichia coli probiotic platform that detects and mitigates oxidative stress in the astronauts’ gut. It senses glycochenodeoxycholic acid (GCDCA), a biomarker indicating food intake, and an oxidative-stress-responsive signal, with both inputs integrated through a split T7 RNA polymerase AND gate to ensure responses only occur when both conditions are present. Its activation triggers the production of trehalose, protecting proteins against oxidative damage, and riboflavin (vitamin B2), supporting detoxification of reactive oxygen species. To enhance storage and deployment, the engineered probiotics are designed to be freeze-dried, improving shelf life and stability for on-demand use. Official Page
