<description>Title: Engineering Barley Stripe Mosaic Virus-Like Particles as Drug Delivery Vectors for Triple Negative Breast Cancer Description: The CSW-UD iGEM team addresses a gap in specific and effective treatments for Triple-Negative Breast Cancer (TNBC) by engineering a novel rod-shaped plant Virus-Like Particle (VLP) platform, based on the Barley Stripe Mosaic Virus (BSMV), into a biocompatible drug carrier to improve the specificity of Doxorubicin, a cancer drug known as the “red devil,” and reduce toxicity. To address this, we modified and engineered the inner channel of BSMV using cysteine mutations to conjugate Doxorubicin with a thiol-Michael reaction, preventing it from indiscriminately attacking healthy cells throughout the body. We further modulated BSMV’s surface to display ligand receptors and endosomal escape peptides to specifically target TNBC and trigger intracellular drug release. We successfully formed a BSMV rod therapy presented with ligands and conjugated with Doxorubicin. Through these efforts, we took meaningful steps towards building a novel drug delivery platform to address the challenges and gaps of current TNBC treatment options.
