        <description>Title: STING - Small TnpB-based Innovative Nuclease toolkit for bacterial Genome editing Description: Genome editing technology is a core approach in synthetic biology. However, traditional CRISPR-Cas9/Cas12 systems encounter difficulties in non-model environmental microorganisms (such as pseudomonads), including high plasmid burden, toxicity caused by heterologous protein expression and low editing efficiency, which severely limits the feasibility of complex genetic engineering. Due to the urgent demand for developing microbial genome-engineering tools, this project focuses on the stringent TAM-recognition constraint of ISHahl1 TnpB, a transposon-derived miniature nuclease, and proposes to broaden its recognition spectrum through semi-rational design guided by structural biology. The aim is to establish an efficient, low-burden miniature editing platform in Pseudomonas oleovorans. Using medium-chain substrate-spectrum expansion of the alkane hydroxylase AlkB and alk-module flux optimization as proof-of-concept applications, this project will systematically verify the precision and practical utility of this editing tool in complex genome manipulation of environmental microorganisms.
