Title: Cocktail Control – An Investigation into the Design of a Methanol Biosensor Description: Methanol poisonings occur as a result of the adulteration of alcoholic beverages, in which methanol is added for profit or during illicit alcohol production. A simple, inexpensive methanol detection test offers a prevention strategy against methanol poisoning. We present two biosensor systems with colorimetric outputs and a hardware sensor design. The systems rely on alcohol oxidase from Phanerochaete chrysosporium (PcAOX) to selectively detect methanol. PcAOX was genetically engineered to enhance its selectivity towards methanol, resulting in seven variants. Molecular simulations showed that four variants exhibited increased selectivity towards methanol relative to WT selectivity. The variants were expressed in Escherichia coli, and the presence of purified proteins was confirmed with Western blotting, although enzymatic activity could not be detected in kinetic assays. An investigation into the reduction of food colorants in a beverage sample using ultrafiltration was conducted. The results suggest that ultrafiltration could be incorporated into the hardware design. Wiki | Official Page
