        <description>Title: Biomanufacturing of 5-Hydroxytryptophan via Multi-Scale Engineering Description: With the continuous prosperity of the global mental health industry, 5-hydroxytryptophan (5-HTP), as a pivotal natural bioactive molecule mediating human neuroregulation and emotional modulation, has witnessed a steady growth in market demand. Leveraging synthetic biology-based microbial cell factory technology, this project achieves the de novo biosynthesis of 5-HTP. Taking Escherichia coli as the chassis cell, this study adopts a multi-dimensional precision engineering strategy. By combining natural strain screening with artificial intelligence-assisted protein design and modification, the tryptophan hydroxylase catalytic module is optimized to enhance enzymatic activity and stability. Meanwhile, the intracellular tetrahydrobiopterin (BH4) synthesis and regeneration pathway is rationally reconstructed, and the metabolic network of the chassis cell is rewired. This research provides an innovative synthetic biology strategy for the eco-friendly, low-cost, and scalable industrial biomanufacturing of 5-HTP.​ Wiki | Official Page</description>
