<description>Title: Primetheus Description: While nuclear genome engineering has advanced rapidly, mitochondrial gene editing remains fundamentally unexplored, current methods being limited to specific single nucleotide substitutions. Prime editing is a gene editing system consisting of a Cas nickase fused to a reverse transcriptase (RT) and a prime editing guide RNA (pegRNA) used as a template to synthesize a new, edited strand without introducing double-strand DNA breaks. Our project Primetheus adapts this system for mitochondria through separate import of the fused Cas-RT complex and the pegRNA into mitochondria. To overcome the technical challenge of verifying intra-mitochondrial RNA delivery, we are also developing a novel, genetically encoded RNA-based readout system to enable continuous live-cell RNA monitoring. In this video we present our progress to date, alongside our modelling, human practices, and education efforts. Primetheus will enable researchers to introduce a wider range of edits, including all base substitutions, as well as longer insertions into mitochondrial DNA, allowing in-depth studies of the mitochondrial genome and unlocking new therapeutic approaches.
