Title: HePatch Description: HePatch is the iGEM 2026 project from iGEM UiOslo-Norway, based at the Department of Biosciences, University of Oslo. Hepatitis B vaccines require storage between 2 and 8 °C from manufacture to administration. Where that cold chain is unreliable, doses are lost and birth-dose coverage falls short. HePatch addresses this by engineering thermostable variants of the hepatitis B surface antigen (HBsAg) and integrating them into dissolvable microneedle patches, with the aim of reducing cold-chain dependency in vaccine delivery. The project combines a computational design pipeline for identifying stabilising mutations with experimental validation in the laboratory, covering cloning, expression and purification, thermostability and antigenicity characterisation, and microneedle formulation. Our Human Practices work has been developed in consultation with ethics committees, regulatory bodies and clinicians working in high-burden settings. This video presents the problem, our approach, and the results obtained during the 2026 season. Wiki | Official Page
