<description>Title: Pigment Green Revolution: Biosynthesis of Indigo from Escherichia coli Description: Indigo connects modern textile manufacturing with centuries of dyeing tradition. Yet conventional production often relies on petrochemical feedstocks and harsh processing conditions. In this video, NNU-Nanjing-China explores how synthetic biology, artificial intelligence, and traditional craftsmanship can inform a different approach. Starting with Escherichia coli, we constructed a TnaA–mFMO biosynthetic pathway and used CRISPR-associated transposase technology to integrate multiple gene copies into the chromosome. To improve intracellular enzyme organization, we developed two complementary AI models: PhaseLACE prioritizes natural intrinsically disordered proteins, while LACE-IDP generates new sequences. Experimental testing identified twelve promising IDP candidates. We then characterized Q97W73-M8 and incorporated an M8–TnaA–mFMO fusion into our engineered chassis. Combining gene dosage optimization with intracellular compartmentalization achieved an indigo titer of 3.199 g/L, 5.52 times that of the original TMF02 strain. Beyond the laboratory, the video follows our dyeing trials, including a water-based workflow and patterned fabrics. Through the iBLUE Cycle, exchanges with researchers, industry partners, craftspeople, and learners helped shape laboratory decisions, practical applications, and educational activities. Safety and inclusivity guided these efforts throughout. Our presentation traces the connections between molecular design, experimental validation, textile applications, and the communities that produce, use, and preserve indigo.
