Title: NemaVOC: A Fungus, a Gas, and a Bacterium Description: NemaVOC is the iGEM 2026 project of the University of Alicante. Plant-parasitic nematodes such as Meloidogyne incognita silently destroy crop roots and are estimated to cost agriculture up to US$157 billion per year, while the chemical nematicides used against them are toxic and are being withdrawn. Our inspiration is Pochonia chlamydosporia, a nematophagous fungus whose volatilome contains 1,4-dimethoxybenzene. We set out to produce its isomer, 1,3-dimethoxybenzene (C5), in Escherichia coli through a three-step polyketide route — ORS with the OAC cyclase, the OrsB decarboxylase and an O-methyltransferase — assembled on two compatible plasmids under a constitutive promoter from the iGEM kit. This video covers the biology of the parasite, why we chose this isomer and this route, our construct design, the molecular docking of C5 against the AlphaFold model of M. incognita acetylcholinesterase, our biosafety approach, and the conversations with growers and the public that shaped the project. We also state openly what is finished and what is still at the bench. Wiki | Official Page
