        <description>Title: Life Factory: Asparaginase Modification - Optimizing Microbial Production of Anti-Leukemia Drugs Description: Acute lymphoblastic leukemia (ALL) is one of the most common cancers in children. Asparaginase (ASNase) is a first-line anti-leukemia drug that inhibits tumor growth by consuming extracellular asparagine. However, current ASNase products derived from natural microorganisms often suffer from deficiencies in production efficiency, stability, and safety, leading to side effects and limited application. This project aims to develop an engineered microbial platform for the efficient production of modified ASNase. We will employ a partitioned rational mutagenesis strategy to modify key accessory residues while maintaining catalytic activity, combining computational prediction and rational design to screen for beneficial mutants. Simultaneously, we will optimize the signal peptide and expression elements to improve enzyme yield and activity. This work provides a synthetic biology-based approach for improving the production of therapeutic enzymes and developing more efficient, stable, and readily available anti-leukemia therapies.
