Title: ROS-Responsive Engineered L. plantarum Delivering Silencing RNA via Membrane Vesicles for Atopic Dermatitis Description: Our presentation video introduces FROST, AFCM-Egypt’s iGEM 2026 project, which explores a synthetic biology approach to atopic dermatitis. It begins by highlighting the challenges patients face with the chronic cycle of itching, skin barrier damage, and inflammation, then explains how FROST aims to address these interconnected problems through a localized, responsive living therapeutic system. The video walks through our engineered Lactiplantibacillus plantarum chassis, the CO-BERA RNA-based therapeutic targeting STAT6 and RelA, and our multifunctional hydrogel designed to support local application and skin barrier protection. It also presents our proposed membrane vesicle-mediated RNA delivery strategy and the biological mechanisms intended to help the therapeutic RNA reach skin cells. Beyond the therapeutic design, the presentation explores our biological containment strategy, which combines environmental sensing and temperature-dependent regulation to help restrict bacterial survival to the intended conditions. It also introduces BOREAS, our integrated modeling framework for investigating storage, adhesion, sensing, RNA-mediated silencing, hydrogel performance, and containment. Finally, the video highlights our human practices, education, inclusivity, and sustainability efforts, demonstrating how stakeholder feedback and community engagement have contributed to shaping our project. Through these interconnected approaches, FROST explores the possibility of a living therapeutic system designed to respond to inflammation, deliver targeted RNA-based therapy, and incorporate safety from the beginning. A treatment that listens first, then acts. Wiki | Official Page
