<description>Title: ORCHID Description: In biomanufacturing, achieving economically viable industrial-scale production requires optimization of titre, rate and yield (TRY). However, engineered genetic constructs often trigger emergent stresses that disrupt cell metabolism and production efficiency. Current methods rely on reactive interventions, adjusting culture conditions or fine-tuning gene expression of biosynthesis pathways after observing the impact of stress, rather than anticipating and preventing these metabolic disruptions. To address this gap, we introduce ORCHID: Observation of Real-time Cellular Health and Intracellular Dynamics For Predictive Biomanufacturing. ORCHID enables real-time monitoring and predictive understanding of cellular physiology through modular multiplexed biosensors designed to simultaneously detect multiple intracellular signals, capturing stress responses across varying culture conditions and gene expression across biosynthetic pathways, linking these cellular dynamics to TRY. To enable predictive biomanufacturing, ORCHID integrates advanced ML/AI to translate the captured high-dimensional data into predictive insights and actionable strategies, advancing understanding of cellular stress and ultimately improving biomanufacturing efficiency and robustness.
