        <description>Title: Sentinel of Light Pollution: Fluorescent Biosensors “Sensing” Light Pollution Description: This project constructs an optogenetic two-hybrid detection system in E. coli to sense light pollution. Red light induces binding of plant phytochrome PhyA with FHY1, recruiting split RNA polymerase subunits to activate luciferase reporter expression. The system includes three plasmids expressing PhyA-FHY1-RNAPα, luciferase, and heme biosynthesis genes. After IPTG induction, bacteria are exposed to 660 nm red light at varying intensities and durations. Chemiluminescence is measured and normalized to cell density. This whole-cell biosensor offers a quantitative, low-cost, and field-deployable tool for monitoring artificial light at night (ALAN), supporting ecological and urban health research.
