<description>Title: RESTART: Rethinking Life After Organ Transplantation Description: RESTART begins with the problem: transplant patients depend on lifelong immunosuppressants, which leave them open to infections and repeated rejection episodes. We then share what patients and clinicians told us, and how their input shaped our goal of improving patients’ quality of life. Next, we introduce the RESTART cascade, a modular AND-gate synNotch circuit that produces IL-10 only when it recognizes both the transplanted organ (HLA-A2) and inflammation (VCAM-1). The video walks through our two-track cell line design, our lab work, and our results, including proof of principle for the full cascade. We also cover our modeling of the receptors and IL-10 diffusion. We close with the road toward a real therapy in regulatory T cells and our outreach on organ transplantation.
