Title: MOSKATEER Description: Malaria continues to impose a disproportionate burden on communities across India and Sub-Saharan Africa, and the rapid spread of insecticide resistance threatens the vector control strategies on which these regions depend. CRISPR-based gene drives offer a transformative approach to mosquito suppression, yet ecological irreversibility, regulatory uncertainty and limited public acceptance remain substantial barriers. MOSKATEER offers a paratransgenic strategy that circumvents these limitations. We engineer symbiotic bacteria to express double-stranded RNA targeting mosGILT, a gene required for Anopheles fertility. A fusion protein comprising tandem MS2 bacteriophage coat protein repeats, a (GGGGS)₄ flexible linker and a functional peptide module packages the dsRNA for stable delivery and RNAi-mediated gene silencing upon ingestion. The platform supports both horizontal spread among adults and vertical inheritance through progeny, amplifying suppression across generations without permanent genomic alteration. Bacteria are immobilised in a sugar matrix baited with acetoin, producing a low-cost, field-deployable sugar trap for resource-limited settings. Wiki | Official Page
