<description>Title: Persephone: PresERvation and Shelf-life Extension of fruits through PostHarvest Optimization by multi-eNgineered Enzymes Description: Persephone is a modular postharvest fruit preservation project targeting ethylene-driven ripening, oxidative damage, and fungal infection. It integrates four modules: ACC deaminase to reduce ethylene biosynthesis, ethylene receptor inhibitory peptides to interfere with ethylene signaling, MnSOD to scavenge superoxide radicals, and chitinase to inhibit fungal growth. ACCD and MnSOD are fused with the cell-penetrating peptide BP100 to improve access to intracellular substrates. Through computational screening and experimental validation, we combined functional components ACCD–BP100, MnSOD–BP100, B03, and chitinase in a chitosan matrix. Under the tested conditions, this formulation delayed banana peel yellowing by approximately three days and showed less visible mold growth than the 1-MCP treatment group. The intended product is a powder containing no live microorganisms, reconstituted before use and applied as a spray within existing postharvest workflows. It complements the cold chain, particularly during long-distance fruit transportation and at stages with insufficient cold-chain coverage.
