Title: Synwave: The Gut-Brain Axis Redefines Alzheimer's Biosensing Description: Dementia is projected to affect 1 in 3 people born in Scotland today, yet Alzheimer’s is often diagnosed too late for disease-modifying drugs to work. The Synwave Screening Test targets detection up to 15 years before symptoms emerge. Leveraging the gut-brain axis, we developed a non-invasive biosensor from Lactobacillus gasseri, which adhered to Caco-2 intestinal epithelial cells with a mean adhesion of 1.7%. We modified a modular receptor platform using the transcription factor CadC, devising an scFv-IC16 receptor that binds Amyloid-β, an Alzheimer’s biomarker, coupling its detection to an amplified reporter signal (BBa_26453DYU). Amyloid-β was expressed in E. coli BL21(DE3) for in vitro testing. A nisin-inducible lysis device (BBa_265W9ZUC) doubles as signal release and biocontainment, ensuring a standardized screening period after which modified bacteria are cleared from the gut. Synwave offers a self-administered, non-invasive and quantitative test read from a stool sample, with potential to enable earlier intervention. Wiki | Official Page
