<description>Title: dSyPhER; Dynamic Synthetic Phosphorylation via Effector Response Description: Diagnostics are episodic by construction: each test is a single observation, and between observations there is no information at all. dSyPhER closes that gap — an engineered cell that detects two disease markers simultaneously, computes an AND decision through a synthetic phosphorylation relay, rejects weak or transient signals with a tunable phosphatase threshold, and reports at the level of translation rather than transcription. Only the sensing layer is disease-specific, so retargeting means swapping a binding domain, not rebuilding the circuit. We demonstrate it on post-remission cancer relapse. The video covers our circuit design and engineering cycles, ODE modelling of the thresholding layer, stakeholder engagement, and our education and public engagement work.
