<description>Title: STING: Small TnpB-based Innovative Nuclease toolkit for bacterial Genome editing Description: STING develops a bacterial genome-engineering toolkit based on the miniature ISHahl1 TnpB nuclease. The system achieved gene deletion in E. coli (4.2–100%) and P. putida (30–44%), alongside transcriptional repression and cytosine base editing. Structure-guided evolution expanded recognition to five noncanonical TAMs. Computational modelling integrated Protenix, Gaussian-mixture scoring, and molecular dynamics simulations to optimize TAM recognition. Integrated Human Practices incorporated feedbacks from experts and stakeholders to improve functionality and biosafety. A self-targeting biocontainment system reduced bacterial survival by up to five orders of magnitude. Core parts include ISHahl1 TnpB (BBa_26F4MI62) and variants with expanded TAM specificities, programmable reRNA, inactive TnpB variants, and base-editing modules, establishing a versatile miniature genome-engineering platform.
