<description>Title: Ciricide: Multitarget cirRNA Pesticide with Minicell Smart Release Description: Rapeseed, one of the world’s three major oil crops, suffers severe yield and quality losses caused by Sclerotinia stem rot. Team SZU-China develops an RNAi biopesticide built around innovatively structured dumbbell-shaped cirRNA. We target three key virulence genes of S. sclerotiorum. With no free termini, this topologically stabilized cirRNA withstands nuclease digestion and achieves much higher environmental stability than linear dsRNA. Engineered E. coli minicells act as smart delivery carriers: they autonomously enrich and encapsulate cirRNA during biosynthesis, protect the payload from enzymatic degradation, and lyse to release cirRNA under the acidic microenvironment secreted by the pathogen. We also develop dedicated cirRNA intelligent design software and promote its industrial application. This reusable platform integrates computational design, molecular engineering, experimental validation and iterative optimization, establishing a complete systematic workflow for developing multi-target cirRNA biopesticides.
