<description>Title: SCENTINEL: A Cell-Free OBP-Based Biosensor for Early Detection of Grapevine Fungal Infections Description: Powdery mildew (Erysiphe necator) threatens global viticulture, driving routine chemical treatments like copper and sulfur that severely contaminate soils. Early detection remains critical, as initial infections are visually undetectable. SCENTINEL is a cell-free, non-GMO biosensing platform designed for early detection of defense-related plant volatile organic compounds (VOCs), primarily methyl salicylate (MeSA). Using our custom Python framework, SCENTINEL CODE, candidate Odorant-Binding Proteins (OBPs) were selected by scoring binding affinity, selectivity, and mixture discrimination. Optimized OBPs were expressed in Escherichia coli, purified via Strep-tag, and immobilized onto nanoporous anodic alumina (NAA) chips. Exposure to MeSA induced a measurable optical shift, providing solid proof of concept for volatile detection. Tailored for winegrower accessibility, SCENTINEL advances precision viticulture to significantly minimize preventative fungicide applications. Wiki | Official Page</description>
