<description>Title: SENTINEL-LOOP: sensing, capture and independent verification of cadmium and lead in water Description: A clear water sample can still carry dissolved cadmium and lead, and a treatment that claims to remove them has to be checked by something other than itself. SENTINEL-LOOP is our design for a closed loop. Two engineered E. coli sensors report cadmium and lead — CadR driving lacZ, and an engineered PbrR driving amilCP. A third construct displays metal-binding domains on the cell surface to capture the metal. Square-wave anodic stripping voltammetry on an in-situ bismuth film then measures what remains, as an independently calibrated method rather than a second opinion from the same biology. This video presents the design, the sequence-verified constructs, the printed hardware with its measured fit and focus tests, and the computational work we used to size the experiments before running them. Although due to time issues, we weren't able to finish everything of our project, we are explicit about the boundary: the sensor, capture and electrochemical characterisation datasets were not completed, every modelled figure is labelled as such, and demonstrating the full loop is the work ahead.
