Inspired by real‑life hot‑pot oil‑splashing pain reported by 719 survey respondents, our project uses de‑novo protein design to engineer two fusion proteins: a capsanthin‑specific pigment‑binding protein and a lipid‑binder‑lipase fusion for oil enrichment & degradation. In our dry‑lab modeling workflow, we combined RFdiffusion3, LigandMPNN, RoseTTAFold3 and AutoDock Vina to screen candidate binders iteratively. In wet‑lab DBTL cycles, we built a low‑cost spectroscopic binding assay and verified that Binder‑5 improved binding performance by ~40 %, and our fusion lipase retained enhanced catalytic activity. We also recorded valuable lessons from terminated lycopene‑related trials regarding measurement‑system stability. Our Human Practices adopted a reusable stakeholder concentric‑circle model plus Discover‑Act‑Analyze‑Feedback loop, shaping our patch toward affordability, safety and inclusive real‑world needs. We also carried out extensive synthetic‑biology education: carnival events, board‑game & card‑game kits, school lectures, reading booklets, debates and multi‑platform science communication, all open‑sourced for the iGEM community. We contribute reproducible pipelines for small‑molecule binder design, low‑cost measurement protocols, and complete HP‑Education frameworks accessible for high‑school iGEM teams.
