Title: PUReCycle——Discovery and Engineering of PURases Description: This presentation video introduces our iGEM project targeting the biological recycling of MDI-based polyether polyurethane waste, a hard-to-degrade plastic that causes severe white pollution. Facing major bottlenecks including scarce high-performance natural urethanases, our work follows the DBTL cycle with two core parts: metagenomic enzyme mining and semi-rational engineering of AbPURase. We built multi-layer computational workflows combining structural prediction, molecular docking and molecular dynamics simulations. Starting from over 30 million metagenomic protein sequences, we screened candidate enzymes and validated them via UPLC wet-lab assays. Through three rounds of mutation engineering, we obtained improved AbPURase mutants balancing catalytic activity and thermal stability. Eight new carbamate-hydrolase candidates were also verified. Besides scientific research, we developed open-source tool SeqPick and a modular reaction controller. We conducted human-practice interviews, academic exchanges and layered synthetic-biology public education. Looking ahead, we aim to advance towards pilot degradation of real PU solid waste, bringing enzymatic plastic recycling from lab to practical production. Wiki | Official Page
