<description>Title: FlaminGone: An Engineered Amyloid Coating for Sustainable Fire Retardancy Description: Traditional chemical fire retardants pose severe health and environmental hazards, creating an urgent need for eco-friendly alternatives. FlaminGone delivers a sustainable bio-coating platform using E. coli’s CsgA curli subunit as a self-assembling amyloid scaffold. Fusing hydrophobic BslA for weatherproofing and polyethylene-binding peptides (PEbp) for substrate anchoring, our Two-Layer System (CsgA-PEbp + CsgA-BslA + APP/EG) overcomes secretion limits and steric hindrance. To accelerate construct engineering, we developed PyroForge, a sequence-only machine learning platform using ESM-2 features to predict protein melting temperature, folding free energy, and mutational stability. Under fire testing, the bio-matrix forms a dense char shield that halts flame propagation and self-extinguishes. BslA prevents rain washout across weathering, while Arabidopsis assays confirm zero ecotoxicity. Swapping binding domains extends this platform to diverse industrial materials.
