Title: A Multicolor Cultural Relic In Situ Restoration System Based on CBM Fusion Enzyme Technology Description: Polychrome artifacts, such as those found in the Beijing Forbidden City, represent an important cultural heritage of human civilization. However, prolonged exposure to environmental factors such as light causes these binding materials to gradually age, leading to pigment fading, powdering, or flaking and detachment. We heterologously expressed key enzymes involved in the biosynthesis of different pigments in Escherichia coli, and employed in vitro enzymatic catalysis to convert cheap and readily available precursors into high-value biopigments. To address the issue of poor photostability, we innovatively fused a cellulose-binding module with the pigment synthases, enabling the produced pigments to be directionally deposited and stably bound to the surface of paper or silk fibers through the anchoring function of the enzymes. This project not only revives ancient colors but also breathes new life into traditional pigments through engineering approaches. Wiki | Official Page
