        <description>Title: Engineering a Programmable Cell Factory for Tunable Pullulan Molecular Weight Description: Pullulan is a natural polysaccharide that is edible, biodegradable, and capable of forming high-quality films, giving it broad potential in food packaging and plant-based capsules. However, its material properties are closely related to molecular weight, which is difficult to regulate flexibly using conventional fermentation methods. Our project aims to transform Aureobasidium melanogenum P16 into a programmable cell factory. At the core of our design is a key transcription factor that regulates three downstream hydrolases associated with changes in pullulan molecular weight. By tuning the expression of this transcription factor, we can influence downstream hydrolase activity, thereby enabling deliberate control over the molecular weight of pullulan. Following the Design–Build–Test–Learn cycle, our project integrates computational modeling, genetic circuit construction, GFP-based characterization, and iterative optimization. We are also developing a portable detection device to support the creation of customizable and sustainable pullulan-based materials for applications in food, packaging, and capsules.
