Title: ProbioRegulon — Small Peptide, Big Potential Description: ProbioRegulon explores the potential of synthetic biology to develop a genetically engineered probiotic platform for delivering a modified version of mcIRBP-19, a peptide associated with research on bitter gourd (Momordica charantia). Our project investigates whether this peptide could influence insulin-receptor signalling and glucose uptake, potentially contributing to complementary approaches to Type 2 diabetes management. We combined computational tools, including AlphaFold2, SignalP, HPEPDOCK, AlphaFold 3 and COPASI, to investigate peptide structure, secretion, predicted receptor interactions and glucose-uptake dynamics. In the laboratory, we transformed three synthetic gene constructs and verified six selected clones, identifying limitations in the original cloning vector. Although computational modelling generated promising hypotheses, peptide expression, secretion and therapeutic effects remain unverified. Through iterative design and stakeholder engagement, we identified priorities for future research. Our long-term concept, Mordilac, envisions a probiotic drink requiring further validation, safety assessment and clinical evaluation. Wiki | Official Page
