To explore a non-surgical alternative, we developed PerioGuard, an engineered probiotic–material hybrid system integrating regenerative protein delivery with enhanced bacterial retention. We engineered Escherichia coli Nissle 1917 (EcN) to produce and secrete CsgA–amelogenin (AMG) under the control of a HucR-based genetic circuit. Using AI-assisted crystal structure prediction, we developed a porous nucleoside-based adsorptive coating (PNAC) assembled from 2FA and cyanuric acid. PNAC promotes bacterial surface attachment, while 2FA enhances biofilm formation and activates CsgA–AMG expression through its metabolic conversion to xanthine. Experiments demonstrated inducible protein expression, PNAC adsorption onto EcN, enhanced biofilm-associated responses, and reduced early macrophage phagocytosis. PerioGuard provides a proof of concept for combining programmable protein delivery with material-assisted bacterial retention, offering a potential strategy for non-surgical periodontal regeneration.
