<description>Title: Engineering Lactiplantibacillus plantarum for Topical Therapy in Psoriasis Description: Psoriasis is a chronic, immune-mediated inflammatory skin disease affecting 2–3% of the global population. At present, there is no complete cure due to its multifactorial and complex nature, with current symptom-focused treatments often limited by variable efficacy and long-term side effects. To address these challenges, we developed a topically applied live biotherapeutic product utiizing bioengineered Lactiplantibacillus plantarum as an alternative treatment option for psoriatic patients. This commensal bacterium acts as a targeted delivery vehicle for fusion peptides combining an IL-23 receptor antagonist and an immunomodulator, effectively blocking the psoriatic inflammatory cascade while regulating skin health. Our project was made possible through extensive stakeholder engagement with patients, clinicians, scientists, industry experts, ethicists, iGEM peers, and the public. Complementing our technical design, we implemented a comprehensive educational campaign aimed at advancing synthetic biology education while actively reducing the social stigma associated with psoriasis.
