Title: AEGIS : Acid-rEsponsive self-assemblinG RGD-ES/KS peptides for tumor Invasion Suppression under weak acid conditions Description: Lung cancer remains the leading cause of cancer death. We engineered a genetically encoded peptide that recognizes the tumor surface and assembles into a physical barrier specifically in the mildly acidic tumor microenvironment (pH ≈ 6.0) by fusing an RGD targeting motif to a pH-responsive polypeptide (ES) scaffold. Through five DBTL cycles, we prioritized the RGD-6ES construct. It demonstrated storage stability, significantly reduced A549 cell viability and migration, reversed EMT markers, and suppressed YAP signaling, while remaining safe to normal bronchial epithelial cells at 0.5 mg/mL. Our Integrated Human Practices refined this trajectory, narrowing the application to NSCLC surgical margins and establishing a preclinical safety package. Addressing dual-use AI risks, we developed PRISM, a locally deployed screening layer. This work delivers a modular, pH-selective platform for tumor microenvironment intervention, with barrier imaging and in vivo validation as next steps. Wiki | Official Page
