<description>Title: Native-like programmablE cartilage Scaffold for Tissue repair (NEST) Description: Cartilage regeneration has long stood as one of the defining challenges of regenerative medicine: true repair requires rebuilding a load-bearing microenvironment where mechanics, anisotropic architecture, and biochemical signals act together. NEST, a native-like programmable engineered cartilage scaffold for tissue repair, uses synthetic biology to manufacture a biomimetic hydrogel that recreates this native logic from molecule to material. Its silk-elastin-like protein backbone is rationally designed to tune mechanical strength, crosslinking behavior, and tissue compatibility, while functional peptide modules introduce cartilage-oriented capabilities such as magnetic alignment, glycosaminoglycan retention, and inflammation-responsive cargo release. A modular ligation system enables these functions to be flexibly assembled and exchanged, tailoring the scaffold to patient-specific needs and diverse clinical conditions. By aligning molecular design with tissue-level function, NEST moves beyond replacing damaged cartilage toward restoring the biological principles that sustain it, envisioning biomaterials as dynamic microenvironments that help tissues recover their native form and function.
