Title: Neurofilux: Early screening of neurodegenerative diseases based on a sandwich antibody assay for NfL protein Description: Neurodegenerative diseases cause irreversible neuronal damage several years before the symptoms appear. Neurofilament light chain (NfL) is a promising blood biomarker, but existing detection methods are either costly or have low sensitivity, making large-scale screening difficult. Therefore, we developed Neurofilux, a low-cost and highly sensitive NfL blood testing system. We identified NfL antigen fragments, designed and screened antibodies using RF antibody, and identified two pairs of highly sensitive antibodies using the sandwich ELISA method. Further, we optimized the design of nanobodies to improve stability and reduce costs. Our future work will iteratively optimize antibody sensitivity through in vitro experiments and adopt digital ELISA technology to achieve a detection sensitivity of up to fg/mL. This detection platform provides an economical and non-invasive solution for the early screening of neurodegenerative diseases. Wiki | Official Page
