<description>Title: EVerse - Reaching Beyond Biological Barriers Description: Biological barriers protect our organs from harmful substances, yet they can also prevent medicines from reaching the tissues that need them. EVerse explores a way around this delivery problem using extracellular vesicles (EVs) and aims to provide researchers with a ready-to-use, non-immunogenic vehicle that can be easily modulated to target and endocytose any cell in the body. An EV-associated scaffold protein presents a SpyCatcher on the surface of the vesicle, which can be conjugated to a SpyTag-equipped nanobody. By exchanging nanobodies, it is possible for the vesicle to recognise different receptors and proteins without rebuilding the entire carrier. We chose the blood-brain barrier (BBB) as our proof of concept to validate the engineered system through cell-based characterisation, transwell assays, and zebrafish embryos. These experiments are an early test of the platform's design and capabilities, with further work needed to establish reliable transport and functional cargo delivery.
