Title: A One-Pot Cancer Detection System via Sequential Thermal Activation Description: Prometheus Fire is a closed-tube, isothermal molecular diagnostic platform developed by Team SRE-China for ultrasensitive detection of microRNA-21. By integrating exponential rolling-circle amplification (E-RCA) with a thermally gated CRISPR-Cas12a system, we achieved temporal separation of signal accumulation and detection. At 30 °C, a programmable ssRNA blocker keeps Cas12a inactive while a zinc-dependent DNAzyme accelerates activator generation. A brief temperature shift to 55–60 °C releases the blocker, triggering rapid fluorescent readout. Key achievements include: Analytical limit of detection of 1.237 fM with high sequence specificity Molecular dynamics and machine-learning-guided optimization of Cas12a and reaction conditions. A portable dual-zone thermal hardware prototype designed for decentralized use Human Practices-driven repositioning from broad screening toward clinically realistic risk triage and post-operative monitoring. Just as Prometheus brought fire to humanity, we aim to bring accessible, high-sensitivity molecular diagnostics closer to the communities that need them most. Wiki | Official Page
