        <description>Title: ReBile：Reprogramming Waste Bile Acids into Therapeutic UDCA Description: Ursodeoxycholic acid (UDCA) is an important therapeutic agent widely used for the treatment of liver and biliary diseases and remains the only FDA-approved drug for primary biliary cholangitis. However, current production methods face significant challenges, including low efficiency, complex chemical synthesis, and ethical concerns associated with traditional bear bile extraction. To address these limitations, we developed a sustainable biosynthesis platform that converts poultry bile waste into high-value UDCA. Using Escherichia coli as a microbial chassis, we engineered a biocatalytic pathway by introducing and optimizing 7α- and 7β-hydroxysteroid dehydrogenases, while enhancing cofactor regeneration to improve conversion efficiency. Computational protein engineering was further applied to optimize key enzymes and maximize production performance. By transforming agricultural by-products into valuable pharmaceuticals, our project combines synthetic biology, enzyme engineering, and circular bioeconomy principles to enable greener UDCA production and promote sustainable waste utilization.
