Our design combines ACC deaminase surface display in Bacillus subtilis, multilayer hydrogel containment, and a proposed cumate-dependent survival-control system. We investigated enzyme expression and activity, tested bacterial containment and cumate release, and developed models to guide protein design, root-zone placement, and inducer delivery. Conversations with stakeholders helped us refine the intended application and identify practical requirements for agricultural use. Our work established component-level evidence and revealed the challenges that must be resolved before integration. Surface-accessible enzyme activity, reproducible containment, and coordinated inducer supply remain key priorities. SoilMate’s next step is to bring these components together and evaluate their performance in plant experiments.
