<description>Title: Reefense Description: Increased ocean temperatures are rapidly accelerating the severity of coral bleaching events worldwide. Thermal stress weakens coral immune defenses, triggers production of toxic reactive oxygen species (ROS), and above 27°C, activates virulence in the pathogen Vibrio coralliilyticus, further harming the coral host. Current treatments struggle to target this pathogen without disrupting the coral holobiont. To address this, our team engineered coral symbiont Bacillus subtilis to secrete Lysdvp001, a phage-derived endolysin that lyses V. coralliilyticus, under a promoter activated by high concentrations of ROS, matching the conditions when the risk of virulence is the highest. Experimental validation demonstrated that our engineered coral probiotic remains comparable to a wild-type environment under low-stress conditions and inhibits V. coralliiyticus when exposed to ROS. Thus, we have created a proactive, biosensing intervention strategy that protects vulnerable coral ecosystems from pathogen-driven bleaching while preserving the natural balance of the coral holobiont.
