<description>Title: BioH₂: Engineering Microbes for Sustainable Hydrogen Production and Byproduct Valorization Description: Hydrogen is a promising clean energy carrier, yet its production remains heavily dependent on fossil fuels. Microbial dark fermentation offers a sustainable alternative, but its potential is constrained by competing metabolic pathways and stringent anaerobic requirements. BioH₂ addresses these challenges by developing Clostridium beijerinckii S63 as an alternative hydrogen-producing chassis. Guided by metabolic modeling, we explored strategies to enhance electron transfer and redirect carbon metabolism. Our engineered strains achieved a hydrogen content of 46.92%, compared with 40.20% in the original strain. Beyond improving hydrogen production, insights from integrated Human Practices and economic analysis encouraged us to rethink the entire production system. We demonstrated that spent fermentation broth could support Bacillus subtilis cultivation, highlighting its potential for value-added microbial biofertilizer production. By integrating metabolic engineering with byproduct valorization, BioH₂ lays the foundation for a circular biomanufacturing platform that aims to make biohydrogen production more resource-efficient, economically viable, and sustainable.
