<description>Title: FORGE: Functional Outcomes via Reprogramming by Genetically Engineered Lactiplantibacillus in Autism Description: SUSTech-MED focuses on the frontier of Autism Spectrum Disorder (ASD) intervention. Our team innovatively utilizes Lactiplantibacillus plantarum L168 as a synthetic biology chassis, engineered to biosynthesize functional molecules—Sulforaphane (SFN) and N-Acetylcysteine (NAC)—while incorporating metabolic pathways to efficiently degrade glutamine and convert neurotoxic brain ammonia into branched-chain amino acids (BCAAs). Delivered via a non-invasive intranasal route to bypass the blood-brain barrier, our system precisely targets the neural microenvironment. By synergistically mitigating ammonia toxicity, modulating neuroinflammation, and alleviating oxidative stress, this platform reshapes the neural microenvironment to relieve core ASD symptoms, pioneering a novel, non-invasive, and promising synthetic biology paradigm for autism care.
