<description>Title: PACERS (PACE with RNA Switch) Description: Duke iGEM explores the broader implications of protein engineering through Phage-Assisted Continuous Evolution (PACE), the challenge of leaky expression in the system, and our project, PACERS (PACE with RNA Switch Control), developed to address this issue. We discuss the design and development of our toehold switch software, mathematical modeling of PACE, and wet-lab experiments to investigate our proposed approach. Additionally, we highlight our stakeholder interviews and the development of a risk assessment tool to evaluate potential risks and guide responsible implementation. Together, these efforts demonstrate our multidisciplinary approach to improving the reliability of continuous protein evolution and exploring its broader applications in medicine, agriculture, and biotechnology.
