Enzyme-based solutions often fail for a simple reason: the substrate fuelling the enzyme runs out. Adding surplus substrate works at lab scale, but with expensive cofactors like ATP it becomes too costly at industrial scale. The alternative is to couple two enzymes, so that one regenerates the spent substrate of the other and creates a near-perpetual catalytic loop. In this audiobook, the iGEM VIT Vellore Dry Lab team walks through the full computational workflow used to characterise such a coupled enzyme system. https://2026.igem.wiki/vit-vellore
