        <description>Title: OMNI-PATCH: Resetting the Microenvironment of Chronic Wounds Description: Chronic wounds impose severe physical and psychological burdens on aging populations, driven by a hostile microenvironment of persistent infection, tissue degradation, and hyper-inflammation. The HKU iGEM team addresses this with Omni-Patch: a smart, biomaterial-driven synthetic biology platform engineered to break the chronic inflammatory cycle and accelerate tissue repair. Crucially, Omni-Patch targets Toll-like Receptor 4 /Myeloid Differentiation factor 2 (TLR4/MD2) complex to rewire the pro-inflammatory environment through inhibiting inflammatory pathways (e.g. NF-κB) while preserving the baseline immune function necessary for natural healing. Our system harnesses cutting-edge computational protein design (RFdiffusion, ProteinMPNN, AlphaFold3, and Molecular Dynamics simulations) to generate novel TLR4/MD2 complex inhibitors, integrating them into a moisture-retaining hydrogel governed by a Nitric Oxide (NO)-responsive delivery mechanism. By deploying therapeutic proteins strictly in response to elevated gaseous NO levels, this innovation delivers precision, on-demand biotherapeutics that revolutionize geriatric wound care.
