Yet current regenerative treatments often rely on complex surgical procedures with limited outcomes. To address this challenge, we developed PerioGuard, a living platform combining engineered probiotics with a functional protective coating. Using the safe probiotic E. coli Nissle 1917, we designed a genetic program that enables the bacteria to produce therapeutic proteins in response to specific signals. To help them survive and function longer, we equipped them with a protective coat that shields them from immune attacks while releasing a signal molecule that promotes bacterial colonization and activates therapeutic protein production. Through this bacteria–material cooperation, PerioGuard promotes alveolar bone regeneration and transforms probiotics into a programmable platform for periodontal repair. From coding genes to rebuilding tissues, PerioGuard brings living therapeutics closer to periodontal regeneration.
