Title: Chariot: Unlocking the Synthetic Utility of OMVs Description: Bacterial outer membrane vesicles (OMVs) are promising drug delivery vectors and vaccine platforms, but their biomanufacturing is limited by low yields, high variability, and costly manufacturing. We present Chariot, an end-to-end platform for the design of vesicle nucleating peptide (VNp)-tagged therapeutics for high-yield OMV loading. Leveraging OMV biofilm penetration and adjuvanticity, we investigate VNp-mediated loading of antimicrobial peptides and bacterial antigens to combat bacterial infections in preventative and acute contexts. Our in silico models, Apollo and Artemis, expand Chariot’s therapeutic applicability through de novo cargo generation and VNp sequence optimization, alongside a custom bioreactor and microfluidic OMV isolation device. In addition to our OMVS (Observe, Modify, Vision, Synthesis) stakeholder framework, we ground our educational initiatives in the MUST (Mutual, Universal, Sustainable, Thoughtful) learning pillars, highlighting meaningful confrontations between diverse communities and the challenges of implementing engineered therapeutics. Chariot enables robust OMV bioengineering for innovative, targeted therapeutic development. Wiki | Official Page
